Product Overview
This rabbit polyclonal antibody recognizes apoptosis-associated speck-like protein containing a CARD (gene Pycard) in mouse samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 21.5 kDa, and the supplier reports an observed band at about 22 kDa.
ASC (PYCARD) is an adaptor protein with a pyrin domain and a CARD. When sensors such as NLRP3, AIM2 or NLRC4 are triggered, ASC polymerizes into a large "speck" that recruits and activates caspase-1, leading to IL-1β and IL-18 maturation and pyroptosis, so it is a standard readout of inflammasome activation.

Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): RM1 cells.
Samples with a confirmed signal (flow cytometry): RAW264.7 cells.
Immunogen
Recombinant mouse ASC/TMS1/Pycard fragment (Lys24–Ser193), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse. UniProt places apoptosis-associated speck-like protein containing a CARD in the cytoplasm, inflammasome, endoplasmic reticulum and mitochondrion. Tissue expression noted by UniProt (mouse): Expressed in small intestine, colon, thymus, spleen, brain, heart, skeletal muscle, kidney, lung and liver. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
