Cellular localization Nucleus. Cytoplasm. Mitochondrion outer membrane. Shuttles between the cytoplasm and the nucleus and heterodimerization with JUN is essential for the nuclear localization. Localization to the cytoplasm is observed under conditions of cellular stress and in disease states. Localizes at the mitochondrial outer membrane in response to genotoxic stress. Phosphorylation at Thr-52 is required for its nuclear localization and negatively regulates its mitochondrial localization. Co- localizes with the MRN complex in the IR-induced foci (IRIF).

Rabbit Monoclonal

Product Overview

Clone IA-1 is a rabbit monoclonal antibody that recognizes cyclic AMP-dependent transcription factor ATF-2 (gene ATF2) in human samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 54.5 kDa, and the supplier reports an observed band at about 70 kDa.

Cyclic AMP-dependent transcription factor ATF-2 (ATF2) is a bZIP transcription factor that binds CRE sites, often as a dimer with c-Jun. It is activated by phosphorylation from stress-activated kinases such as p38 and JNK, and it is studied in stress responses, DNA damage signaling and cancer.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:30

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human HUVEC cells, human K562 cells, human HEL cells, human SiHa cells, human SH-SY5Y cells, human Caco-2 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Samples with confirmed staining (IHC): human colon cancer tissue, human stomach cancer tissue, mouse brain tissue, rat brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with confirmed staining (ICC/IF): HeLa cells.

Immunogen

Synthetic peptide based on the sequence of human ATF2.

Reactivity Notes

The supplier lists reactivity with human. UniProt places cyclic AMP-dependent transcription factor ATF-2 in the nucleus, cytoplasm and mitochondrion outer membrane. Tissue expression noted by UniProt (human): Ubiquitously expressed, with more abundant expression in the brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Immunoprecipitation 1:30. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 54.5 kDa; observed band at about 70 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human ATF2.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
