Vendor description (summary): Boster Bio Anti-ATG4A Antibody Picoband® catalog # A06539-2.

Biological background

Biological context: Cysteine protease required for the cytoplasm to vacuole transport (Cvt) and autophagy. Cleaves the C-terminal amino acid of ATG8 family proteins to reveal a C-terminal glycine. Exposure of the glycine at the C-terminus is essential for ATG8 proteins conjugation to phosphatidylethanolamine (PE) and insertion to membranes, which is necessary for autophagy. Preferred substrate is GABARAPL2 followed by MAP1LC3A and GABARAP. Has also an activity of delipidating enzyme for the PE-conjugated forms.

Expression and localization notes: cellular localization: Cytoplasm., tissue context: Widely expressed, at a low level, and the highest expression is observed in skeletal muscle and brain. Also detected in fetal liver..

Common research applications

  • Western blotting (WB): Compare ATG4A levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of ATG4A in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify ATG4A-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Cysteine protease ATG4A is an enzyme that in humans is encoded by the ATG4A gene. It is mapped to Xq22.3. Autophagy is the process by which endogenous proteins and damaged organelles are destroyed intracellularly. Autophagy is postulated to be essential for cell homeostasis and cell remodeling during differentiation, metamorphosis, non-apoptotic cell death, and aging. Reduced levels of autophagy have been described in some malignant tumors, and a role for autophagy in controlling the unregulated cell growth linked to cancer has been proposed. This gene encodes a member of the autophagin protein family. The encoded protein is also designated as a member of the C-54 family of cysteine proteases.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cytoplasm.
  • Tissue details: Widely expressed, at a low level, and the highest expression is observed in skeletal muscle and brain. Also detected in fetal liver.
  • Research category: Signal Transduction
