Product Overview

This rabbit polyclonal antibody detects ubiquitin-like modifier-activating enzyme ATG7 in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 78.0 kDa, and the supplier reports an observed band at about 78 kDa.

ATG7 is the E1-like activating enzyme of both ubiquitin-like conjugation systems in autophagy. It activates ATG12 for transfer to ATG5 and activates LC3 family proteins for transfer to ATG3, making it essential for autophagosome formation and a common target in autophagy knockout studies.

Samples with a confirmed band (WB): human HepG2 cells, human PC-3 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human colorectal adenocarcinoma tissue, human liver cancer tissue, human lung adenocarcinoma tissue, human meningioma tissue, human ovarian serous adenocarcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with confirmed staining (ICC/IF): HeLa cells.

Samples with a confirmed signal (flow cytometry): HepG2 cells.

Recombinant human ATG7 fragment (Asp71–Lys645), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 92.9%.

Reactivity Notes

The supplier lists reactivity with human. UniProt places ubiquitin-like modifier-activating enzyme ATG7 in the cytoplasm and preautophagosomal structure. Tissue expression noted by UniProt (human): Widely expressed, especially in kidney, liver, lymph nodes and bone marrow. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
