Product Overview
This rabbit polyclonal antibody detects ATP-citrate synthase (gene ACLY) in human, mouse and rat samples and is validated for flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 120.8 kDa, and the supplier reports an observed band at about 125 kDa.
ATP-citrate lyase (ACLY) splits citrate into acetyl-CoA and oxaloacetate in the cytosol. This supplies acetyl-CoA for fatty acid and cholesterol synthesis and for histone acetylation, and ACLY is the target of the cholesterol-lowering drug bempedoic acid.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A594 cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human ATP citrate lyase fragment (Met1–Ile180), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 95%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ATP-citrate synthase in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
