Product Overview
Clone CCG-2 is a rabbit monoclonal antibody that recognizes serine/threonine-protein kinase B-raf (gene BRAF) in human, mouse and rat samples and is validated for Western blot, IHC, immunoprecipitation and flow cytometry. The predicted molecular weight is 84.4 kDa, and the supplier reports an observed band at about 85 kDa.
B-Raf (BRAF) is a RAF family serine/threonine kinase activated by RAS that phosphorylates MEK1/2 (MAP2K1/2), which in turn activate ERK in the MAPK pathway controlling cell growth and differentiation. The V600E mutation constitutively activates BRAF and is common in melanoma, thyroid and colorectal cancers, making BRAF a major drug target.

Samples with confirmed staining (IHC): human ovarian cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human B-Raf.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places serine/threonine-protein kinase B-raf in the nucleus, cytoplasm and cell membrane. Tissue expression noted by UniProt (human): Brain and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
