Product Overview

This rabbit polyclonal antibody targets beta-secretase 1 (gene BACE1) in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 55.8 kDa, and the supplier reports an observed band at about 70 kDa.

β-secretase 1 (BACE1) is a membrane-bound aspartic protease that performs the first cleavage of the amyloid precursor protein (APP), which, followed by γ-secretase cleavage, generates amyloid-β peptides. Because of this role, BACE1 is a major subject of Alzheimer′s disease research; it also processes other substrates, including neuregulin-1.

Samples with a confirmed band (WB): human HeLa cells, human U-87MG cells, human SH-SY5Y cells, rat brain tissue, mouse brain tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Recombinant human BACE1 fragment (Ala185–Glu432), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places beta-secretase 1 in the cell membrane, Golgi apparatus, endoplasmic reticulum and endosome. Tissue expression noted by UniProt (human): Expressed at high levels in the brain and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
