Product Overview
Clone CHC-2 is a rabbit monoclonal antibody that recognizes bcl2-associated agonist of cell death (gene BAD) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 18.4 kDa, and the supplier reports an observed band at about 23 kDa.
BAD is a pro-apoptotic BH3-only member of the Bcl-2 family. When dephosphorylated it binds Bcl-2 and Bcl-xL and frees BAX and BAK to trigger apoptosis, while survival signals such as AKT phosphorylate BAD and sequester it with 14-3-3 proteins. It is widely used to study growth factor-dependent cell survival.

Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, human placenta tissue, mouse small intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human bad.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places bcl2-associated agonist of cell death in the mitochondrion outer membrane and cytoplasm. Tissue expression noted by UniProt (human): Expressed in a wide variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
