Product Overview
Clone CHI-2 is a rabbit monoclonal antibody that recognizes bcl-2 homologous antagonist/killer (gene BAK1) in human and mouse samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 23.4 kDa, and the supplier reports an observed band at about 23–25 kDa.
BAK (BAK1) is a pro-apoptotic Bcl-2 family effector that resides in the mitochondrial outer membrane. After activation by BH3-only proteins, it oligomerizes to form pores that release cytochrome c and other apoptogenic factors, working alongside BAX in the intrinsic apoptosis pathway.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): mouse liver tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human bak.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places bcl-2 homologous antagonist/killer in the mitochondrion outer membrane. Tissue expression noted by UniProt (human): Expressed in a wide variety of tissues, with highest levels in the heart and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
