Biological background
Protein function (datasheet): Promotes cell survival. Blocks dexamethasone-induced apoptosis. Mediates survival of postmitotic Sertoli cells by suppressing death-promoting activity of BAX. .
Scientific background (datasheet): Bcl-2-like protein 2 is a protein that in humans is encoded by the BCL2L2 gene. This gene encodes a pro-survival (anti-apoptotic) member of the bcl-2 protein family. Its gene is mapped to human chromosome 14q11.2-q12. In all, it has got 193 amino acids. Generally speaking, the BCL2 can be found in brain, colon and salivary gland. Acting as the anti- and pro-apoptotic regulators, it has played a vital role in reducing the cell apoptosis under cytotoxic conditions.
Cellular localization (datasheet): Mitochondrion membrane ; Peripheral membrane protein . Loosely associated with the mitochondrial membrane in healthy cells. During apoptosis, tightly bound to the membrane.
Tissue details (datasheet): Expressed (at protein level) in a wide range of tissues with highest levels in brain, spinal cord, testis, pancreas, heart, spleen and mammary glands. Moderate levels found in thymus, ovary and small intestine. Not detected in salivary gland, muscle or liver. Also expressed in cell lines of myeloid, fibroblast and epithelial origin. Not detected in most lymphoid cell lines. .
Sequence similarities (datasheet): Belongs to the Bcl-2 family.
Research relevance and current trends
- Commonly studied in contexts related to Apoptosis,Cancer,Cell Biology,Intracellular,Oncoproteins,Oncoproteins/Suppressors.
- Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
- Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).
Common research applications
- Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
- ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
- Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
- Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
- Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.
Notes for experimental interpretation
- Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
- Cross-reactivity (datasheet): No cross-reactivity with other proteins
- Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.
As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
