Product Overview
Clone AAOH-2 is a rabbit monoclonal antibody that recognizes apoptosis regulator Bcl-2 (gene BCL2) in human and mouse samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 26.3 kDa, and the supplier reports an observed band at about 26 kDa.
Bcl-2 (BCL2) is an anti-apoptotic protein located mainly on the mitochondrial outer membrane that binds and restrains pro-apoptotic Bcl-2 family members such as BAX, BAK and BH3-only proteins. It was first identified through the t(14;18) translocation in follicular lymphoma, and it is a central regulator of mitochondrial apoptosis and a target of BH3-mimetic drugs.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human lung adenocarcinoma tissue, human thyroid cancer tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human bcl-2.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places apoptosis regulator Bcl-2 in the mitochondrion outer membrane, nuclear membrane, endoplasmic reticulum membrane and cytoplasm. Tissue expression noted by UniProt (human): Expressed in a variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
