Product Overview
Clone AFAG-2 is a rabbit monoclonal antibody that recognizes B2 bradykinin receptor (gene BDKRB2) in human and mouse samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 44.5 kDa, and the supplier reports an observed band at about 78 kDa.
The B2 bradykinin receptor (BDKRB2) is a constitutively expressed G protein-coupled receptor for bradykinin and kallidin. It signals through Gq, phospholipase C and Ca2+ and mediates vasodilation, vascular permeability, pain and inflammation. BDKRB2 is the target of icatibant, which is used to treat hereditary angioedema attacks.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
| Immunoprecipitation | 1:50 |
Samples with a confirmed band (WB): human HeLa cells, human 293T cells, human MCF-7 cells, human K562 cells, mouse lung tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human liver cancer tissue, mouse small intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human BDKRB2.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places B2 bradykinin receptor in the cell membrane. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
