Biological background

Biological context: Iron-trafficking protein involved in multiple processes such as apoptosis, innate immunity and renal development. Binds iron through association with 2,5-dihydroxybenzoic acid (2,5- DHBA), a siderophore that shares structural similarities with bacterial enterobactin, and delivers or removes iron from the cell, depending on the context. Iron-bound form (holo-24p3) is internalized following binding to the SLC22A17 (24p3R) receptor, leading to release of iron and subsequent increase of intracellular iron concentration. In contrast, association of the iron-free form (apo-24p3) with the SLC22A17 (24p3R) receptor is followed by association with an intracellular siderophore, iron chelation and iron transfer to the extracellular medium, thereby reducing intracellular iron concentration. Involved in apoptosis due to interleukin-3 (IL3) deprivation: iron-loaded form increases intracellular iron concentration without promoting apoptosis, while iron-free form decreases intracellular iron levels, inducing expression of the proapoptotic protein BCL2L11/BIM, resulting in apoptosis. Involved in innate immunity, possibly by sequestrating iron, leading to limit bacterial growth. .

Expression and localization notes: cellular localization: Secreted . Upon binding to the SLC22A17 (24p3R) receptor, it is internalized., tissue context: Detected in lung, spleen, uterus, vagina and epididymis. ..

Common research applications

  • Western blotting (WB): Compare B2M levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of B2M in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify B2M-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Beta-2 microglobulin also known as B2M is a component of MHC class I molecules, which are present on all nucleated cells (excludes red blood cells). In humans, the beta-2-microglobulin protein is encoded by the B2M gene. The protein has a predominantly beta-pleated sheet structure that can form amyloid fibrils in some pathological conditions. The encoded antimicrobial protein displays antibacterial activity in amniotic fluid. A mutation in this gene has been shown to result in hypercatabolic hypoproteinemia.
  • Cross reactivity: No cross-reactivity with other proteins
  • Cellular localization: Secreted . Upon binding to the SLC22A17 (24p3R) receptor, it is internalized.
  • Tissue details: Detected in lung, spleen, uterus, vagina and epididymis. .
  • Research category: Immunoglobulins,Immunology

We have observed staining in human plasma. What should we do? Is anti-Beta 2 Microglobulin/B2M antibody supposed to stain plasma positively?
From what I have seen in literature plasma does express B2M. From what I have seen in Uniprot.org, B2M is expressed in leukocyte, peripheral blood, cervix carcinoma, dendritic cell, leukemia, trachea, retina skin, plasma, urine, tear, liver, macrophage, among other tissues. Regarding which tissues have B2M expression, here are a few articles citing expression in various tissues: Liver, Pubmed ID: 24275569 Macrophage, Pubmed ID: 25356553 Peripheral blood, Pubmed ID: 16108498 Plasma, Pubmed ID: 8084451 Retina, and Skin, Pubmed ID: 15489334 Tear, Pubmed ID: 25946035 Trachea, Pubmed ID: 14702039 Urine, Pubmed ID: 7554280
