Product Overview
Clone BF-1 is a rabbit monoclonal antibody that recognizes actin, cytoplasmic 1 (gene ACTB) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 41.7 kDa, and the supplier reports an observed band at about 42 kDa.
Beta-actin (ACTB) is a cytoplasmic actin found in nearly all cells that assembles into filaments forming the cytoskeleton. It supports cell shape, migration and division and also acts in the nucleus in gene regulation, and it is widely used as a loading control in Western blot.

Samples with a confirmed band (WB): human GES-1 cells, human HeLa cells, human A431 cells, human K562 cells, rat C6 cells, rat PC-12 cells, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:5000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human heart, mouse testis, human bladder cancer, mouse heart tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human beta-actin.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places actin, cytoplasmic 1 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
