Product Overview

Clone AAGC-2 is a rabbit monoclonal antibody that recognizes BH3-interacting domain death agonist (gene BID) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 22.0 kDa, and the supplier reports an observed band at about 22 kDa.

BID is a BH3-only pro-apoptotic Bcl-2 family protein that links death receptor signaling to mitochondria. After cleavage by caspase-8 (or other proteases), truncated BID (tBID) moves to mitochondria and activates BAX and BAK, causing cytochrome c release and amplifying the apoptotic signal.

Samples with a confirmed band (WB): Jurkat cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human lung tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Samples with confirmed staining (ICC/IF): Jurkat cells.

Immunogen

Synthetic peptide based on the sequence of human bid.

Reactivity Notes

The supplier lists reactivity with human. UniProt places BH3-interacting domain death agonist in the cytoplasm, mitochondrion membrane and mitochondrion outer membrane. Tissue expression noted by UniProt (human): Expressed in spleen, pancreas and placenta (at protein level)... The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:10. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on primate samples?
A.Reactivity with primate samples has not been tested by the supplier. Aligning the immunogen sequence with the primate ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 22.0 kDa; observed band at about 22 kDa.
