Product Overview
This rabbit polyclonal antibody recognizes B-cell linker protein (gene BLNK) in human, mouse and rat samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 50.5 kDa, and the supplier reports an observed band at about 70 kDa.
BLNK (also called SLP-65) is a linker protein phosphorylated by SYK after B-cell receptor engagement. Its phosphotyrosines recruit BTK, PLCγ2, VAV and GRB2, coupling the receptor to calcium, MAPK and NF-κB signaling. BLNK is essential for normal B-cell development, and its loss causes a form of agammaglobulinemia.

Samples with a confirmed band (WB): human Raji cells, human Jurkat cells, rat kidney tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): T47D cells.

Recombinant human BLNK fragment (Asp59–Ser456), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places B-cell linker protein in the cytoplasm and cell membrane. Tissue expression noted by UniProt (human): Expressed in B-cell lineage and fibroblast cell lines (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
