Product Overview

Clone IHE-2 is a rabbit monoclonal antibody that recognizes polycomb complex protein BMI-1 (gene BMI1) in human and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 36.9 kDa, and the supplier reports an observed band at about 40 kDa.

BMI1 is a core component of the Polycomb repressive complex 1 (PRC1), where it stimulates RING1B-mediated monoubiquitination of histone H2A at lysine 119 to keep target genes silent. It represses the INK4A/ARF locus and is required for self-renewal of hematopoietic, neural and other stem cells, and it is studied in stem cell biology and cancer.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human K562 cells, human U2OS cells, human A431 cells, human A549 cells, rat PC-12 cells, rat C6 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human bmi1.

Reactivity Notes

The supplier lists reactivity with human and rat. UniProt places polycomb complex protein BMI-1 in the nucleus and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
