Product Overview
Clone BBB-2 is a rabbit monoclonal antibody that recognizes bone morphogenetic protein 4 (gene BMP4) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 46.6 kDa, and the supplier reports an observed band at about 47 kDa.
Bone morphogenetic protein 4 (BMP4) is a TGF-β superfamily growth factor that activates SMAD1/5/8 through BMP receptors. It patterns the early embryo, including ventral mesoderm and epidermal fates, and regulates bone, kidney, lung and tooth development, and it is widely used to direct stem cell differentiation in culture.

Samples with a confirmed band (WB): human HeLa cells, human COLO320 cells, human Caco-2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

The supplier lists reactivity with human. Tissue expression noted by UniProt (human): Expressed in the lung and lower levels seen in the kidney. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
