Product Overview
This rabbit polyclonal antibody recognizes breast cancer type 1 susceptibility protein (gene BRCA1) in human, mouse and rat samples and is validated for ICC/IF and Western blot. The predicted molecular weight is 207.7 kDa, and the supplier reports an observed band at about 290 kDa.
BRCA1 is a tumor suppressor that forms an E3 ubiquitin ligase with BARD1 and acts in several nuclear complexes that sense and repair DNA double-strand breaks, especially by homologous recombination. It also supports cell cycle checkpoints and genome stability, and inherited BRCA1 mutations greatly increase the risk of breast and ovarian cancer.

Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells, human T47D cells, human Caco-2 cells, rat C6 cells, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Immunogen
Recombinant human BRCA1 fragment (Thr1681–Glu1781), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 73.3% and 74.3%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places breast cancer type 1 susceptibility protein in the nucleus, chromosome and cytoplasm. Tissue expression noted by UniProt (human): Isoform 1 and isoform 3 are widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
