Product Overview
This rabbit polyclonal antibody detects lys-63-specific deubiquitinase BRCC36 in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 36.1 kDa, and the supplier reports an observed band at about 36 kDa.
BRCC36 (BRCC3) is a JAMM-family metalloprotease deubiquitinase that specifically cleaves Lys63-linked ubiquitin chains. It is the catalytic subunit of the nuclear BRCA1-A complex, which regulates ubiquitin signaling at DNA double-strand breaks, and of the cytoplasmic BRISC complex, which acts in inflammasome and interferon receptor signaling.

Samples with a confirmed band (WB): human HEK293 cells, rat kidney tissue, rat pancreas tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): U87 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places lys-63-specific deubiquitinase BRCC36 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
