Product Overview
Clone DBH-2 is a rabbit monoclonal antibody that recognizes bromodomain-containing protein 4 (gene BRD4) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 152.2 kDa, and the supplier reports an observed band at about 240 kDa.
BRD4 is a BET family chromatin reader whose two bromodomains bind acetylated histones. It recruits the P-TEFb complex to promote transcriptional elongation by RNA polymerase II and stays on chromatin during mitosis to help transmit gene expression programs. BRD4 is concentrated at super-enhancers of oncogenes such as MYC and is the main target of BET inhibitors like JQ1.

Samples with a confirmed band (WB): :mouse brain tissue, human Jurkat cells, human HepG2 cells, human MCF-7 cells, human PC-12 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human kidney tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human brd4.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places bromodomain-containing protein 4 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Ubiquitously expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
