Overview

This antibody is intended for detection of POU4F1 (Zinc finger protein Helios) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-BRN3A/POU4F1 Antibody Picoband® catalog # A05403-1. Tested in ELISA, WB applications. This antibody reacts with Human. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E. coli-derived human BRN3A recombinant protein (Position: R311-Y419). (reported region: R311-Y419).
  • Molecular weight context: reported MW: 45 kDa; calculated MW: 39411 MW
  • Reactivity: Human
  • Applications: ELISA, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Zinc finger protein Helios; POU class 4 homeobox 1. POU domain, class 4, transcription factor 1 (POU4F1), also known as brain-specific homeobox/POU domain protein 3A (BRN3A), homeobox/POU domain protein RDC-1 or Oct-T1, is a protein that in humans is encoded by the POU4F1 gene. It is mapped to 13q31.1. This gene encodes a member of the POU-IV class of neural transcription factors. This protein is expressed in a subset of retinal ganglion cells and may be involved in the developing sensory nervous system. This protein may also promote the growth of cervical tumors. A translocation of this gene is associated with some adult acute myeloid leukemias. Functional note: Multifunctional transcription factor with different regions mediating its different effects. Acts by binding (via its C-terminal domain) to sequences related to the consensus octamer motif 5'-ATGCAAAT-3' in the regulatory regions of its target genes. Regulates the expression of specific genes involved in differentiation and survival within a subset of neuronal lineages. It has been shown that activation of some of these genes requires its N-terminal domain, maybe through a neuronal-specific cofactor. Ativates BCL2 expression and protects neuronal cells from apoptosis (via the N-terminal domain). Induces neuronal process outgrowth and the coordinate expression of genes encoding synaptic proteins. Exerts its major developmental effects in somatosensory neurons and in brainstem nuclei involved in motor control. Stimulates the binding affinity of the nuclear estrogene receptor ESR1 to DNA estrogen response element (ERE), and hence modulates ESR1-induced transcriptional activity. May positively regulate POU4F2 and POU4F3. Regulates dorsal root ganglion sensory neuron specification and axonal projection into the spinal cord. Plays a role in TNFSF11-mediated terminal osteoclast differentiation. Negatively regulates its own expression interacting ly with a highly conserved autoregulatory domain surrounding the transcription initiation site. Reported localization: Nucleus. Expression/tissue context: Expressed in the brain and the retina. Present in the developing brain, spinal cord and eye.

Common research applications

  • Western blotting: compare relative POU4F1 (Zinc finger protein Helios) levels across conditions; band patterns may reflect isoforms and processing.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

We have observed staining in human retina. Do you have any suggestions? Is anti-BRN3A/POU4F1 antibody supposed to stain retina positively?
From what I have seen in literature retina does express POU4F1. From what I have seen in Uniprot.org, POU4F1 is expressed in secondary oocyte, placenta, retina, among other tissues. Regarding which tissues have POU4F1 expression, here are a few articles citing expression in various tissues: Placenta, Pubmed ID: 1357630 Retina, Pubmed ID: 7623109, 15057823
