Product Overview

This rabbit polyclonal antibody detects protein BTG2 in human samples and is validated for flow cytometry and Western blot. The predicted molecular weight is 17.4 kDa, and the supplier reports an observed band at about 22 kDa.

BTG2 (also called TIS21 or PC3) is an antiproliferative protein of the BTG/Tob family induced by p53 after DNA damage. It interacts with the CCR4–NOT deadenylase complex to promote mRNA decay, and it helps control cell cycle exit and neuronal differentiation.

Validated Applications

Western blot 0.1–0.5 µg/mL
Flow cytometry (fixed cells) 1–3 µg/1×106 cells

Samples with a confirmed band (WB): human HepG2 cells, human PC-3 cells, human U2OS cells, human A549 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with a confirmed signal (flow cytometry): A431 cells.

Immunogen

Synthetic peptide from the N-terminal region of human BTG2. Sequence identity with the mouse and rat orthologs: 88.9%.

Reactivity Notes

The supplier lists reactivity with human. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL; Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on primate samples?
A.Reactivity with primate samples has not been tested by the supplier. Aligning the immunogen sequence with the primate ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 17.4 kDa; observed band at about 22 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human BTG2. Sequence identity with the mouse and rat orthologs: 88.9%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
