Product Overview
This rabbit polyclonal antibody detects mitotic checkpoint serine/threonine-protein kinase BUB1 beta (gene BUB1B) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 119.5 kDa, and the supplier reports an observed band at about 120–130 kDa.
BubR1 (BUB1B) is a kinase-family protein of the spindle assembly checkpoint. As part of the mitotic checkpoint complex with MAD2, BUB3 and CDC20, it holds the anaphase-promoting complex (APC/C) inactive until every chromosome is correctly attached to the spindle, and it also helps recruit the motor CENPE to kinetochores. It is studied in mitosis, chromosome instability, aneuploidy and aging.

Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse testis tissue, rat testis tissue, human colon cancer tissue, human mammary cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): U87 cells.
Immunogen
Recombinant human BubR1/BUB1B fragment (Lys26–Glu448), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places mitotic checkpoint serine/threonine-protein kinase BUB1 beta in the cytoplasm, nucleus and chromosome. Tissue expression noted by UniProt (human): Highly expressed in thymus followed by spleen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
