Cellular localization Nucleus. Endoplasmic reticulum. Cytoplasm, cytosol. In quiescent cells, present in very small amounts in the cytosol. Following induction of cell growth, first localizes to the endoplasmic reticulum and only later to the nucleus. Localization at the endoplasmic reticulum requires dephosphorylation at Tyr-10 and Tyr-30.

Rabbit Monoclonal

Product Overview

Clone IIO-6 is a rabbit monoclonal antibody that recognizes protein c-Fos in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 40.7 kDa, and the supplier reports an observed band at about 41 kDa.

c-Fos is a transcription factor that pairs with JUN family proteins to form the AP-1 complex, which binds DNA and regulates genes controlling proliferation, differentiation and survival. FOS is a classic immediate early gene rapidly induced by growth factors, stress and neuronal activity, so c-Fos expression is widely used as a marker of activated neurons.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human HeLa cells, human RT4 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human c-Fos.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places protein c-Fos in the nucleus, endoplasmic reticulum and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
