Product Overview
This rabbit polyclonal antibody recognizes complement C5 in rat samples and is validated for IHC and Western blot. The predicted molecular weight is 9.0 kDa, and the supplier reports an observed band at about 115 kDa.
Complement C5 is cleaved by C5 convertases from all complement pathways into C5a and C5b. C5a is a potent anaphylatoxin and chemoattractant for neutrophils and other leukocytes, while C5b starts assembly of the membrane attack complex with C6–C9. It is widely studied in inflammation, infection and complement-mediated disease.

Samples with a confirmed band (WB): rat liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTAbuffer (pH 8.0).
Immunogen
Recombinant rat C5a fragment (Asp1–Arg77), expressed in E. coli. Sequence identity with the human and mouse orthologs: 62% and 82%, respectively.
Reactivity Notes
The supplier lists reactivity with rat. UniProt describes complement C5 as secreted and also located in the target cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
