Anti-CACNA1G (CaV3.1) Antibody (#ACC-021) is a highly specific antibody directed against an epitope of the rat CaV3.1 channel. The antibody can be used in western blot, immunocytochemistry, and immunohistochemistry applications. It has been designed to recognize CACNA1G from human, rat, and mouse samples. Anti-CACNA1G (CaV3.1)-ATTO Fluor-594 Antibody (#ACC-021-AR) is directly labeled with ATTO-594 fluorescent dye. ATTO dyes are characterized by strong absorption (high extinction coefficient), high fluorescence quantum yield, and high photo-stability. The ATTO-594 fluorescent label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594. Anti-CACNA1G (CaV3.1)-ATTO Fluor-594 Antibody is ideal for multiplex staining studies.

Target CACNA1G

Overview

Anti-CACNA1G (CaV3.1)-ATTO Fluor-594 Antibody is an antibody targeting Voltage-dependent T-type calcium channel subunit α1G Polyclonal raised in Rabbit (ATTO-594. Maximum absorption 601 nm; Maximum fluorescence 627 nm. The fluorescence is excited most efficiently in the 580 - 615 nm range. This label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594.). This antibody is commonly used in IF, IHC to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Voltage-dependent T-type calcium channel subunit α1G (also reported as Voltage-dependent T-type calcium channel subunit α1G).
  • Immunogen/epitope region: Intracellular, N-terminus.
  • Homology note: Mouse - identical; human - 20/22 amino acid residues identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • KO-validated: yes (validation context may be assay-dependent).
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #ACC-021), and immunohistochemistry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

Voltage-dependent Ca2+ channels provide a pathway for rapid influx of Ca2+ into cells, which plays a crucial role in both electrical and metabolic signaling.1 T-type currents are transduced via channel proteins encoded by three genes that compose a subfamily within the CaV channel family.2-3 The activity of T-type channels contributes to several known physiological and pathophysiological phenomena including burst firing in neurons, pacemaking activity in the heart and secretion from endocrine tissues.2 There are three cloned T-type channel isoforms. CACNA1G (CaV3.1) and CACNA1H (CaV3.2) are widely distributed whereas the expression of CACNA1I (CaV3.3) is restricted to the central nervous system.2CACNA1G and CACNA1H are also expressed in the kidney, but little is known about their physiological role there.

Research relevance and current trends

  • Mapping receptor/channel localization across neuronal subtypes and subcellular compartments.
  • Linking trafficking or surface expression changes to activity-dependent signaling and plasticity.
  • Using KO/KD or blocking-peptide concepts to strengthen antibody-based target assignment.

Common research applications

  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
  • Immunofluorescence/ICC: assess subcellular localization and co-localization with markers in cells or sections.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Conceptual control: KO/KD samples provide orthogonal support for target assignment when available.
  • Provided control suggestions: Negative control: BLP-CC021.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
