Product Overview

This rabbit polyclonal antibody targets multifunctional protein CAD in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and ELISA. The predicted molecular weight is 243.0 kDa, and the supplier reports an observed band at about 260 kDa.

CAD is a large multifunctional enzyme that performs the first three steps of de novo pyrimidine synthesis: carbamoyl phosphate synthetase II, aspartate transcarbamylase and dihydroorotase. Because growing cells need pyrimidines for DNA and RNA synthesis, CAD is regulated by mTORC1 and MAPK signaling and is studied in cell growth, cancer metabolism and nucleotide biosynthesis.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human, Rat)
IHC (paraffin sections) 2–5 µg/mL (Human, Mouse, Rat)
Immunocytochemistry / Immunofluorescence 5 µg/mL (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human 293T cells, human HeLa cells, rat C6 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human gastric lymphoma tissue, human lung adenocarcinoma tissue, human prostate cancer tissue, human spleen tissue, human squamous carcinoma tissue, mouse testis tissue, rat testis tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with confirmed staining (ICC/IF): MCF-7 cells.

Immunogen

Recombinant human CAD fragment (Arg1403–Asp2198), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places multifunctional protein CAD in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human, Rat); IHC (paraffin sections) 2–5 µg/mL (Human, Mouse, Rat); Immunocytochemistry / Immunofluorescence 5 µg/mL (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.

A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 243.0 kDa; observed band at about 260 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human CAD fragment (Arg1403–Asp2198), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
