Overview

This antibody is intended for detection of CALB1 in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Calbindin/CALB1 Antibody Picoband® catalog # A03047. Tested in ELISA, Flow Cytometry, IF, IHC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E. coli-derived human Calbindin recombinant protein (Position: A2-N261). (reported region: A2-N261).
  • Molecular weight context: reported MW: 28 kDa; calculated MW: 30,025 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: ELISA, Flow Cytometry, IF, IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

calbindin 1, 28kDa. Calbindin is a calcium-binding protein belonging to the troponin C superfamily. And it mapped to 8q21.3. Calretinin is expressed in central and peripheral nervous system and in many normal and pathological tissues. The rat and human calretinin exhibit 98% sequence homology and 91% homology to many other species. Two calcium binding proteins, calbindin and calretinin, have been reported to be expressed in abundance in Purkinje cells and other cell types in the cerebellum. Functional note: Buffers cytosolic calcium. May stimulate a membrane Ca (2+)-ATPase and a 3',5'-cyclic nucleotide phosphodiesterase. Reported localization: Hemidesmosome, Membrane, Single-pass type II membrane protein, Basement membrane. Expression/tissue context: Detected in skin. In the cornea, it is detected in the epithelial basement membrane, the epithelial cells, and at a lower level in stromal cells. Stratified squamous epithelia. Found in hemidesmosomes. Expressed in cornea, oral mucosa, esophagus, intestine, kidney collecting ducts, ureter, bladder, urethra and thymus but is absent in lung, blood vessels, skeletal muscle and nerves.

Research relevance and current trends

  • Calcium Binding Proteins: Researchers commonly examine how CALB1 relates to this theme using model systems and orthogonal readouts.
  • Calcium Signaling: Researchers commonly examine how CALB1 relates to this theme using model systems and orthogonal readouts.
  • Cell Type Marker: Researchers commonly examine how CALB1 relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative CALB1 levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of CALB1 across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
