Product Overview
This rabbit polyclonal antibody detects calreticulin in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 48.1 kDa, and the supplier reports an observed band at about 60 kDa.
Calreticulin (CALR) is an abundant Ca2+-binding lectin chaperone of the endoplasmic reticulum. Together with calnexin, it folds and quality-checks newly made glycoproteins, and it also stores ER calcium and helps load peptides onto MHC class I. Calreticulin exposed on the cell surface acts as an "eat me" signal, and CALR mutations drive some myeloproliferative neoplasms.

Samples with a confirmed band (WB): human placenta tissue, human U-87MG cells, human U-937 cells, human HepG2 cells, human THP-1 cells, human T-47D cells, human PC-3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human intestinal cancer tissue, human prostatic cancer tissue, human thyroid cancer tissue, mouse intestine tissue, rat intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with a confirmed signal (flow cytometry): HepG2 cells.

Recombinant human Calreticulin/CALR fragment (Glu18–Ser323), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places calreticulin in the endoplasmic reticulum lumen, cytoplasm, cell surface and sarcoplasmic reticulum lumen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
