Product Overview

Clone AAFF-3 is a rabbit monoclonal antibody that recognizes calcium/calmodulin-dependent protein kinase type II subunit alpha (gene CAMK2A) in human, mouse and rat samples and is validated for Western blot, IHC, immunoprecipitation and flow cytometry. The predicted molecular weight is 54.1 kDa, and the supplier reports an observed band at about 54 kDa.

CaMKIIα (CAMK2A) is the neuron-enriched alpha subunit of calcium/calmodulin-dependent protein kinase II, a dodecameric holoenzyme. After Ca2+/calmodulin binding and autophosphorylation at Thr286, it stays active on its own, which allows it to support long-term potentiation at excitatory synapses. It is a classic target in studies of learning, memory and synaptic plasticity.

Samples with a confirmed band (WB): rat hippocampus tissue, rat brain tissue, rat C6 cells, mouse brain tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human colon cancer tissue, rat brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
