Biological background

CAMK2B (protein: T-cell surface glycoprotein CD3 zeta chain) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Calcium/calmodulin-dependent protein kinase that functions autonomously after Ca (2+)/calmodulin-binding and autophosphorylation, and is involved in dendritic spine and synapse formation, neuronal plasticity and regulation of sarcoplasmic reticulum Ca (2+) transport in skeletal muscle. In neurons, plays an essential structural role in the reorganization of the actin cytoskeleton during plasticity by binding and bundling actin filaments in a kinase-independent manner. This structural function is required for correct targeting of CaMK2A, which acts downstream of NMDAR to promote dendritic spine and synapse formation and maintain synaptic plasticity which enables long-term potentiation (LTP) and hippocampus-dependent learning. In developing hippocampal neurons, promotes arborization of the dendritic tree and in mature neurons, promotes dendritic remodeling. Participates in the modulation of skeletal muscle function in response to exercise. In slow-twitch muscles, is involved in regulation of sarcoplasmic reticulum (SR) Ca (2+) transport and in fast-twitch muscle participates in the control of Ca (2+) release from the SR through phosphorylation of triadin, a ryanodine receptor-coupling factor, and phospholamban (PLN/PLB), an endogenous inhibitor of SERCA2A/ATP2A2. . Reported cellular localization context: Cytoplasm, cytoskeleton . Cytoplasm, cytoskeleton, microtubule organizing center, centrosome . Sarcoplasmic reticulum membrane ; Peripheral membrane protein ; Cytoplasmic side . In slow- twitch muscle, evenly distributed between longitudinal SR and junctional SR. Tissue expression notes (as provided): Widely expressed. Expressed in adult and fetal brain. Expression is slightly lower in fetal brain. Expressed in skeletal muscle. .

Research relevance and current trends

  • Research context keywords from the source record include: Calcium Signaling,Calmodulin Pathway,Cardiovascular,Hypertrophy,Neuroscience,Neurotransmission,Protein Phosphorylation,Ser/Thr Kinases,Signal Transduction,Signaling Pathway.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.

Workflow ideas (metafield): Validate CAMK2B antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect CAMK2B expression by Western blot in cell or tissue lysates, Detect CAMK2B in FFPE tissue sections by immunohistochemistry, Localize CAMK2B by immunofluorescence/immunocytochemistry in cultured cells

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 75 kDa; calculated MW: 72678 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.
