Product Overview

Clone ABBF-3 is a rabbit monoclonal antibody that recognizes calcium/calmodulin-dependent protein kinase type II subunit delta (gene CAMK2D) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 56.4 kDa, and the supplier reports an observed band at about 50 kDa.

CaMKIIδ (CAMK2D) is the main CaMKII isoform in the heart. It phosphorylates ion channels, phospholamban and ryanodine receptors to tune Ca2+ handling and excitation–contraction coupling in cardiomyocytes. Excess CaMKIIδ activity is linked to cardiac hypertrophy, heart failure and arrhythmia, making it a major cardiac research target.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence Not specified by the supplier

Samples with a confirmed band (WB): human Daudi cells, human U-87MG cells, human HUVEC cells, human A549 cells, rat C6 cells, mouse MH-S cells, mouse Neuro-2a cells, mouse RAW264.7 cells, mouse lung tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Samples with confirmed staining (ICC/IF): mouse MH-S cells.

Immunogen

Synthetic peptide based on the sequence of human caMKII delta.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places calcium/calmodulin-dependent protein kinase type II subunit delta in the cell membrane and sarcoplasmic reticulum membrane. Tissue expression noted by UniProt (human): Expressed in cardiac muscle and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence (dilution not specified). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 56.4 kDa; observed band at about 50 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human caMKII delta.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
