Product Overview

This rabbit polyclonal antibody recognizes collagen alpha-2(IV) chain (gene COL4A2) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 167.6 kDa, and the supplier reports an observed band at about 24–35 kDa.

Collagen α2(IV) chain (COL4A2) pairs with COL4A1 to form the most widespread type IV collagen network of basement membranes. Its C-terminal fragment canstatin inhibits angiogenesis, and COL4A2 is studied in vascular integrity, basement membrane biology and cerebral hemorrhage.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)
IHC (paraffin sections) 2–5 µg/mL (Human)

Samples with a confirmed band (WB): human placenta tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human breast cancer tissue, human placenta tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide from the C-terminal region of human canstatin. The peptide sequence is conserved in rat and mouse.

Reactivity Notes

The supplier lists reactivity with human. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human); IHC (paraffin sections) 2–5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 167.6 kDa; observed bands at about 24 and 35 kDa. The observed band is much smaller than the calculated size, which can indicate a processed form or a shorter isoform; check the supplier images.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human canstatin. The peptide sequence is conserved in rat and mouse.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
