Product Overview

This rabbit polyclonal antibody recognizes troponin T, cardiac muscle (gene TNNT2) in human, mouse and rat samples and is validated for ELISA and Western blot. The predicted molecular weight is 35.9 kDa, and the supplier reports an observed band at about 43 kDa.

Cardiac troponin T (cTnT, TNNT2) is the subunit that anchors the troponin complex to tropomyosin on the thin filament of heart muscle. Together with troponin I and C, it makes cardiac contraction depend on calcium. It is a widely used blood biomarker of myocardial injury, and TNNT2 mutations are a known cause of inherited cardiomyopathies.

Validated Applications

Western blot 0.1–0.5 µg/mL
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): rat heart tissue, mouse heart tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Immunogen

Recombinant human Cardiac Troponin T fragment (Lys88–Lys298), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Heart. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL; ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 35.9 kDa; observed band at about 43 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Cardiac Troponin T fragment (Lys88–Lys298), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
