Product Overview

This rabbit polyclonal antibody targets caspase-14 (gene CASP14) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 27.7 kDa, and the supplier reports an observed band at about 28 kDa.

Caspase-14 is a non-apoptotic caspase expressed mainly in differentiating keratinocytes of the epidermis. It is involved in cornification and helps process filaggrin into natural moisturizing factors, supporting skin barrier function and hydration. It is studied in epidermal differentiation and skin barrier disorders.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL (Human)
Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): rat brain tissue, rat liver tissue, rat spleen tissue, A431 cells, NIH3T3 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human tonsil tissue, human oesophagus squama cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide from the C-terminal region of human caspase-14. It differs from the rat and mouse sequences at four residues.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places caspase-14 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Expressed in keratinocytes of adult skin suprabasal layers (from spinous layers to the stratum granulosum and stratum corneum) (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 0.5–1 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on primate samples?
A.Reactivity with primate samples has not been tested by the supplier. Aligning the immunogen sequence with the primate ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 27.7 kDa; observed band at about 28 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human caspase-14. It differs from the rat and mouse sequences at four residues.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
