Product Overview

Clone 8B6 is a mouse monoclonal antibody that recognizes caspase-3 (gene CASP3) in human samples and is validated for flow cytometry, IHC and Western blot. The predicted molecular weight is 31.6 kDa, and the supplier reports an observed band at about 35 kDa.

Caspase-3 is the major executioner caspase of apoptosis. Activated by initiator caspases such as caspase-8 and caspase-9, it cleaves hundreds of cellular proteins, including PARP1, leading to the typical features of apoptotic cell death. Cleaved (active) caspase-3 is one of the most widely used apoptosis markers in Western blot and immunohistochemistry.

Validated Applications

Western blot 0.1–0.5 µg/mL
IHC (paraffin sections) 0.5–1 µg/mL
Flow cytometry (fixed cells) 1–3 µg/1×106 cells

Samples with a confirmed band (WB): human placenta tissue, human K562 cells, human HepG2 cells, human Raji cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): human tonsil tissue, human intestinal cancer tissue, human placenta tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with a confirmed signal (flow cytometry): HepG2 cells.

Immunogen

Recombinant human Caspase-3 fragment (Thr67–Asp175), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 86% and 90%, respectively.

Reactivity Notes

The supplier lists reactivity with human. UniProt places caspase-3 in the cytoplasm. Tissue expression noted by UniProt (human): Highly expressed in lung, spleen, heart, liver and kidney. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL; IHC (paraffin sections) 0.5–1 µg/mL; Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 31.6 kDa; observed band at about 35 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Caspase-3 fragment (Thr67–Asp175), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 86% and 90%, respectively.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
