Product Overview
This rabbit polyclonal antibody detects caspase-4 (gene CASP4) in human samples and is validated for ELISA, flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 43.3 kDa, and the supplier reports an observed band at about 48 kDa.
Caspase-4 is a human inflammatory caspase that directly senses cytosolic lipopolysaccharide from Gram-negative bacteria. Binding LPS activates it to cleave gasdermin D, triggering pyroptosis through the non-canonical inflammasome and indirectly activating NLRP3. It is studied in innate immunity, sepsis and ER stress, and its mouse functional counterpart is caspase-11.

Samples with a confirmed band (WB): human HeLa cells, human COLO-320 cells, human SW620 cells, human HepG2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, human mammary cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): SiHa cells, PC-3 cells.
Samples with confirmed staining (ICC/IF): human tonsil tissue.
Immunogen
Recombinant human Caspase 4 fragment (His111–Arg194), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places caspase-4 in the cytoplasm, endoplasmic reticulum membrane and mitochondrion. Tissue expression noted by UniProt (human): Widely expressed, including in keratinocytes and colonic and small intestinal epithelial cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
