Product Overview

This rabbit polyclonal antibody targets caspase-4 (gene CASP4) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 43.3 kDa, and the supplier reports an observed band at about 20 kDa.

Caspase-4 is a human inflammatory caspase that directly senses cytosolic lipopolysaccharide from Gram-negative bacteria. Binding LPS activates it to cleave gasdermin D, triggering pyroptosis through the non-canonical inflammasome and indirectly activating NLRP3. It is studied in innate immunity, sepsis and ER stress, and its mouse functional counterpart is caspase-11.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL (Human)
Western blot 0.1–0.5 µg/mL (Human)

Samples with a confirmed band (WB): MCF-7 cells, HeLa cells, Jurkat cells, CEM cells, SW620 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human mammary cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide from the N-terminal region of human caspase 4.

Reactivity Notes

The supplier lists reactivity with human. UniProt places caspase-4 in the cytoplasm, endoplasmic reticulum membrane and mitochondrion. Tissue expression noted by UniProt (human): Widely expressed, including in keratinocytes and colonic and small intestinal epithelial cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human); IHC (paraffin sections) 0.5–1 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 43.3 kDa; observed band at about 20 kDa. The observed band is much smaller than the calculated size, which can indicate a processed form or a shorter isoform; check the supplier images.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human caspase 4.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
