Product Overview
Clone EEG-3 is a rabbit monoclonal antibody that recognizes caspase-8 (gene CASP8) in human samples and is validated for Western blot. The predicted molecular weight is 55.4 kDa, and the supplier reports an observed band at about 50 kDa.
Caspase-8 is the initiator caspase of the extrinsic apoptosis pathway. Recruited through FADD to death receptors such as Fas and TNFR1, it becomes active and cleaves caspase-3, caspase-7 and BID. Caspase-8 also restrains necroptosis by cleaving RIPK1 and RIPK3, which makes it a central switch between apoptosis, necroptosis and inflammation.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|
Samples with a confirmed band (WB): human Jurkat cells, human Caco-2 cells, human K562 cells, human RT4 cells, human A549 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human caspase-8.
Reactivity Notes
The supplier lists reactivity with human. UniProt places caspase-8 in the cytoplasm, nucleus and cell projection. Tissue expression noted by UniProt (human): Isoform 1, isoform 5 and isoform 7 are expressed in a wide variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
