Product Overview
Clone GAI-3 is a rabbit monoclonal antibody that recognizes caspase-8 (gene CASP8) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 55.4 kDa, and the supplier reports an observed band at about 55 kDa.
Caspase-8 is the initiator caspase of the extrinsic apoptosis pathway. Recruited through FADD to death receptors such as Fas and TNFR1, it becomes active and cleaves caspase-3, caspase-7 and BID. Caspase-8 also restrains necroptosis by cleaving RIPK1 and RIPK3, which makes it a central switch between apoptosis, necroptosis and inflammation.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places caspase-8 in the cytoplasm, nucleus and cell projection. Tissue expression noted by UniProt (human): Isoform 1, isoform 5 and isoform 7 are expressed in a wide variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
