Product Overview
This rabbit polyclonal antibody recognizes caspase-8 (gene CASP8) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 55.4 kDa, and the supplier reports an observed band at about 45 kDa.
Caspase-8 is the initiator caspase of the extrinsic apoptosis pathway. Recruited through FADD to death receptors such as Fas and TNFR1, it becomes active and cleaves caspase-3, caspase-7 and BID. Caspase-8 also restrains necroptosis by cleaving RIPK1 and RIPK3, which makes it a central switch between apoptosis, necroptosis and inflammation.

Validated Applications
| IHC (paraffin sections) | 0.5–1 µg/mL (Human) |
|---|---|
| Western blot | 0.1–0.5 µg/mL (Human) |
Samples with a confirmed band (WB): HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Samples with confirmed staining (IHC): human mammary cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide from the C-terminal region of human caspase-8(P10).
Reactivity Notes
The supplier lists reactivity with human. UniProt places caspase-8 in the cytoplasm, nucleus and cell projection. Tissue expression noted by UniProt (human): Isoform 1, isoform 5 and isoform 7 are expressed in a wide variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
