Product Overview

This rabbit polyclonal antibody detects cathepsin D (gene Ctsd) in mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 45.0 kDa, and the supplier reports an observed band at about 45 kDa.

Cathepsin D (CTSD) is a lysosomal aspartic protease that carries out bulk breakdown of proteins in acidic compartments. It also participates in processing of proteins such as amyloid precursor protein. CTSD is studied in lysosomal function, neurodegeneration and breast cancer, where its secretion has been widely investigated.

Validated Applications

Western blot 0.1–0.5 µg/mL (Mouse)
IHC (paraffin sections) 0.5–1 µg/mL (Mouse, Rat)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Mouse)

Samples with a confirmed band (WB): mouse liver tissue, mouse brain tissue, mouse thymus tissue, NEURO cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): mouse kidney tissue, mouse liver tissue, rat liver tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Samples with a confirmed signal (flow cytometry): mouse liver tissue.

Immunogen

Recombinant mouse Cathepsin D fragment (Glu65–Leu410), expressed in E. coli. Sequence identity with the human and rat orthologs: 85% and 91%, respectively.

Reactivity Notes

The supplier lists reactivity with mouse and rat. UniProt describes cathepsin D as secreted and also located in the lysosome and melanosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Mouse); IHC (paraffin sections) 0.5–1 µg/mL (Mouse, Rat); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Mouse). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on bovine samples?
A.Reactivity with bovine samples has not been tested by the supplier. Aligning the immunogen sequence with the bovine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 45.0 kDa; observed band at about 45 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant mouse Cathepsin D fragment (Glu65–Leu410), expressed in E. coli. Sequence identity with the human and rat orthologs: 85% and 91%, respectively.
