Product Overview

Clone 31C05 is a rabbit monoclonal antibody that recognizes pro-cathepsin H (gene CTSH) in human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 37.4 kDa, and the supplier reports an observed band at about 28 kDa.

Cathepsin H (CTSH) is a lysosomal cysteine protease that works mainly as an aminopeptidase, removing single amino acids from protein ends, and also as an endopeptidase. It contributes to overall protein breakdown in lysosomes and to processing of lung surfactant protein B, and it is studied in lysosomal biology and immune regulation.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human RT4 cells, human A431 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human lung cancer tissue, human liver cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human cathepsin H.

Reactivity Notes

The supplier lists reactivity with human. UniProt places pro-cathepsin H in the lysosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 37.4 kDa; observed band at about 28 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human cathepsin H.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
