Key elements and design rationale

  • Antibody format: Mouse Monoclonal Mouse IgG2a
  • Clone number: Clone: 12C7
  • Immunogen / epitope context: E.coli-derived human Caveolin-1 recombinant protein (Position: G4-I178). Human Caveolin-1 shares 95% and 94% amino acid (aa) sequence identity with mouse and rat Caveolin-1, respectively. (reported region: G4-I178).
  • Molecular weight context: reported MW: 22 kDa; calculated MW: 46659 MW
  • Reactivity: Human
  • Applications: IF, IHC, IHC-F, WB

As a monoclonal antibody, the reagent targets a defined epitope, supporting consistency across experiments; epitope masking by PTMs or conformational changes can affect signal.

Biological background

Isocitrate dehydrogenase [NADP] cytoplasmic; caveolin 1, caveolae protein, 22kDa. CAV1 ( Caveolin-1) is a protein that in humans is encoded by the CAV1 gene. The CAV1 gene is mapped to 7q31.2. The scaffolding protein encoded by this gene is the main component of the caveolae plasma membranes found in most cell types. The protein links integrin subunits to the tyrosine kinase FYN, an initiating step in coupling integrins to the Ras-ERK pathway and promoting cell cycle progression. The gene is a tumor suppressor gene candidate and a negative regulator of the Ras-p42/44 MAP kinase cascade. CAV1 and CAV2 are located next to each other on chromosome 7 and express colocalizing proteins that form a stable hetero-oligomeric complex. By using alternative initiation codons in the same reading frame, two isoforms (alpha and beta) are encoded by a single transcript from this gene. Functional note: May act as a scaffolding protein within caveolar membranes. Interacts ly with G-protein alpha subunits and can functionally regulate their activity. Involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Its binding to DPP4 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. Recruits CTNNB1 to caveolar membranes and may regulate CTNNB1-mediated signaling through the Wnt pathway. Negatively regulates TGFB1-mediated activation of SMAD2/3 by mediating the internalization of TGFBR1 from membrane rafts leading to its subsequent degradation. Mediates the recruitment of CAVIN proteins (CAVIN1/2/3/4) to the caveolae. Reported localization: Cell membrane. Peripheral membrane protein. Golgi apparatus membrane. Peripheral membrane protein. Trans-Golgi network. Caveola. Peripheral membrane protein. Membrane raft. Expression/tissue context: Skeletal muscle, liver, stomach, lung, kidney and heart (at protein level). Expressed in the brain.

Research relevance and current trends

  • Calcium Channels: Researchers commonly examine how CAV1 (Isocitrate dehydrogenase [NADP] cytoplasmic) relates to this theme using model systems and orthogonal readouts.
  • Calcium Signaling: Researchers commonly examine how CAV1 (Isocitrate dehydrogenase [NADP] cytoplasmic) relates to this theme using model systems and orthogonal readouts.
  • Cancer: Researchers commonly examine how CAV1 (Isocitrate dehydrogenase [NADP] cytoplasmic) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative CAV1 (Isocitrate dehydrogenase [NADP] cytoplasmic) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of CAV1 (Isocitrate dehydrogenase [NADP] cytoplasmic) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

My question regarding product M00179-1, anti-Caveolin-1/CAV1 antibody (monoclonal, 12C7). I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
It is not recommended storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M00179-1 anti-Caveolin-1/CAV1 antibody (monoclonal, 12C7), we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
We need to test anti-Caveolin-1/CAV1 antibody (monoclonal, 12C7) M00179-1 on human adipose tissue for research purposes, then I may be interested in using anti-Caveolin-1/CAV1 antibody (monoclonal, 12C7) M00179-1 for diagnostic purposes as well. Is the antibody suitable for diagnostic purposes?
The products we sell, including anti-Caveolin-1/CAV1 antibody (monoclonal, 12C7) M00179-1, are only intended for research use. They would not be suitable for use in diagnostic work. If you have the means to develop a product into diagnostic use, and are interested in collaborating with us and develop our product into an IVD product, please contact us for more discussions.
