Cellular localization Cytoplasm, cytosol . Nucleus. Endoplasmic reticulum. Mitochondrion . Mainly localizes in the cytosol. Co-localizes with SYT11 in neutrites. Co-localizes with SNCAIP in brainstem Lewy bodies. Mitochondrial localization gradually increases with cellular growth. Also relocates to dysfunctional mitochondria that have lost the mitochondrial membrane potential; recruitment to mitochondria is PINK1- dependent.

Rabbit Monoclonal

Product Overview

Clone 20C55 is a rabbit monoclonal antibody that recognizes cystathionine beta-synthase (gene CBS) in human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 60.6 kDa, and the supplier reports an observed band at about 61 kDa.

Cystathionine β-synthase (CBS) is a pyridoxal phosphate-dependent enzyme that catalyzes the first step of the transsulfuration pathway, joining homocysteine and serine to form cystathionine. This removes homocysteine and supplies cysteine, and CBS also produces hydrogen sulfide. Inherited CBS deficiency is the most common cause of classical homocystinuria.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): MCF-7 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human CBS.

Reactivity Notes

The supplier lists reactivity with human. UniProt places cystathionine beta-synthase in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): In the adult strongly expressed in liver and pancreas, some expression in heart and brain, weak expression in lung and kidney. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
