Product Overview

Clone 23C16 is a rabbit monoclonal antibody that recognizes E3 SUMO-protein ligase CBX4 in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 61.4 kDa, and the supplier reports an observed band at about 50 kDa.

CBX4 (Pc2) is a Polycomb group protein of the PRC1 complex that recognizes methylated histone H3 through its chromodomain. It is also an E3 SUMO ligase that sumoylates proteins such as CtBP and proteins involved in DNA repair. CBX4 is studied in gene silencing, stem cell maintenance and the DNA damage response.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): human 293T cells, human Jurkat cells, human HepG2 cells, human K562 cells, rat C6 cells, mouse kidney tissue, mouse RAW264.7 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
