Product Overview
This rabbit polyclonal antibody targets integrin alpha-M (gene ITGAM) in human, mouse and rat samples and is validated for flow cytometry, IHC and Western blot. The predicted molecular weight is 127.2 kDa, and the supplier reports an observed band at about 45 kDa.
Integrin alpha-M (ITGAM, CD11b) pairs with integrin beta-2 to form Mac-1, also called complement receptor 3. It binds the complement fragment iC3b, fibrinogen and ICAM-1, supporting adhesion, migration and phagocytosis by monocytes, macrophages and neutrophils. CD11b is one of the most widely used myeloid cell markers.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human tonsil tissue, mouse spleen tissue, rat spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): THP-1 cells.
Immunogen
Recombinant human CD11b fragment (Phe17–Thr382), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 79% and 61%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places integrin alpha-M in the cell membrane and membrane raft. Tissue expression noted by UniProt (human): Predominantly expressed in monocytes and granulocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
