Biological background

Integrin alpha-M; Integrin alpha-M. Integrin alpha M (ITGAM) is one protein subunit that forms the heterodimeric integrin alpha-M beta-2 (αMβ2) molecule, also known as macrophage-1 antigen (Mac-1) or cluster of differentiation molecule 11B (CD11B). It is mapped to 16p11.2. ITGAM has a role in vascular repair after mechanical arterial injury. It is implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement-coated particles. What's more, ITGAM probably recognizes the R-G-D peptide in C3b, and it is also a receptor for fibrinogen, factor X and ICAM1. Functional note: Integrin alpha-M/beta-2 is implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement-coated particles. It is identical with CR-3, the receptor for the iC3b fragment of the third complement component. It probably recognizes the R-G-D peptide in C3b. Integrin alpha-M/beta-2 is also a receptor for fibrinogen, factor X and ICAM1. It recognizes P1 and P2 peptides of fibrinogen gamma chain. Alpha-M/beta-2 play a critical role in mast cell development and in immune complex-mediated glomerulonephritis. Mice expressing a null mutation of the alpha-M subunit gene demonstrate increase in neutrophil accumulation, in response to a impaired degranulation and phagocytosis, events that apparently accelerate apoptosis in neutrophils. We have observed staining in mouse blood. Any tips? Is anti-CD11b/ITGAM antibody supposed to stain blood positively?
According to literature blood does express ITGAM. According to Uniprot.org, ITGAM is expressed in bone marrow cell, blood, liver, among other tissues. Regarding which tissues have ITGAM expression, here are a few articles citing expression in various tissues: Blood, Pubmed ID: 1346576, 3539202 Liver, Pubmed ID: 19159218
We are currently using anti-CD11b/ITGAM antibody A00144 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says mouse. Is it possible that the antibody can work on zebrafish tissues as well?
The anti-CD11b/ITGAM antibody (A00144) has not been validated for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity.
