Product Overview
This rabbit polyclonal antibody targets receptor-type tyrosine-protein kinase FLT3 in human samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 112.9 kDa, and the supplier reports an observed band at about 160 kDa.
FLT3 (CD135) is a receptor tyrosine kinase activated by FLT3 ligand on hematopoietic progenitor cells and dendritic cell precursors. It drives their survival, proliferation and differentiation through RAS–MAPK, PI3K–AKT and STAT5 signaling. Activating FLT3 mutations, such as internal tandem duplications, are among the most common alterations in acute myeloid leukemia.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human hccp tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human spleen tissue, human cerebellum tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human CD135/FLT3 fragment (Ala66–Ser993), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places receptor-type tyrosine-protein kinase FLT3 in the membrane and endoplasmic reticulum lumen. Tissue expression noted by UniProt (human): Detected in bone marrow, in hematopoietic stem cells, in myeloid progenitor cells and in granulocyte/macrophage progenitor cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
